I am an illustrator and a designer, who loves to make funny art. I specialize in comic and cartoon design. I am a daddy, a husband, a friend, a fan, a geek, and a musician.We wish to further the research in our studies on the regulation of cell proliferation and differentiation in normal and neoplastic cells by the bcl-2 proto-oncogene product, Bcl-2. During the previous period of support we have identified the basic properties and functions of Bcl-2 and determined its function as a regulator of growth and proliferation. We have found that Bcl-2 has complex effects on the growth and death of cells; it may protect against apoptosis and induce cell proliferation in different cell types. Bcl-2 also plays an essential role in terminal differentiation of lymphoid cells. This suggests a direct role of Bcl-2 in apoptosis-independent differentiation of lymphoid cells. In fact, we found that Bcl-2 prevents the development of plasma cells in vitro. Bcl-2 is expressed in all stages of plasmacytoma differentiation, but is lost in plasma cell neoplasms. Therefore, we propose to further study the role of Bcl-2 in terminal differentiation and apoptosis-independent death of lymphoid cells. We have already mapped the protein region of Bcl-2 that is involved in its interaction with anti-apoptotic proteins and we plan to study how interaction of Bcl-2 with specific proteins regulates its function. We also plan to continue our studies on the role of the pro-apoptotic molecule, Bax, in lymphoid cell apoptosis. We have found that Bax, in contrast to the anti-apoptotic Bcl-2, can induce apoptosis in a cell-type specific fashion; its function is regulated by the BH3 motif. We will now further study the regulation of this motif in Bax and its role in apoptosis and differentiation. We have recently made an interesting finding concerning the role of Bcl-2 in the development of tumors in the immune system.
